How did an American Company, Eli Lilly use the knowledge of r-DNA technology to produce human insulin?


In 1983, American company Eli Lily prepared human insulin by using the knowledge of r-DNA technology in the following steps-


a) DNA corresponding to A and B chain is prepared separately.


b) Plasmid bacteria are extracted.


c) Inserting DNA corresponding to A and B chain separately in plasmid.


d) Transforming the bacteria with this recombinant plasmid.


e) Desired product i.e. insulin is expressed from this DNA.


f) A and B chains are purified


g) They are linked together with disulphide bonds which makes them act like human insulin.
