Mention the role of (i) selectable marker, (ii) Ori and (iii) rop in E. coli cloning vector pBR322.

(i) pBR322 has two antibiotic-resistant genes for ampicillin and tetracycline, which acts as a selectable marker which is used for differentiation between the recombinants and non-recombinants. Thus, it helps in the identification and the selection of the transformants.


(ii) Ori is the origin of replication site in cloning vector pBR322, which is a genetic sequence which acts as an initiation site for replication of DNA.


(iii) ROP in pBR322 is known as the repressor of primer, which is a protein which plays the role of regulating the copy number.
