What was the challenge for production of insulin using rDNA techniques ? How did Eli Lilly produce insulin using rDNA technology?

The challenge for the production of insulin using rDNA techniques was the assembling of insulin components into mature insulin form.


Eli Lilly produces insulin using rDNA technology by:


(i) Taking two DNA sequences corresponding to A and B chains of human insulin.


(ii) They introduced them in plasmids of E.coli to produce insulin chains. These Chains were produced separately, extracted, and combined by creating disulphide bonds to form human insulin.


(iii) Insulin in the human pancreas is synthesized as a pro-hormone containing the C peptide, which is removed in the mature hormone.


(iv) The insulin synthesized by recombinant DNA technology did not contain C peptide and was prepared in the mature form.
